The first filly to accomplish this was Cleopatra (1917). She finished 3rd in the 1920 Kentucky Derby; her son, Pompey finished 5th in the 1926 Kentucky Derby.

Correction. Cleopatra finished 15th, not 3rd.

I'm not sure why she didn't come up in my first search, but Ascension (1872) finished 10th in the 1875 Kentucky Derby; her son Ascender finished 4th in the 1883 Derby.
